Yes. Spectare fills slots via JavaScript after the page loads, so it is unaffected by page caching. WP Rocket, W3 Total Cache, WP Super Cache, and LiteSpeed Cache all work without any configuration.
The one thing to check: make sure your caching plugin is not set to defer or exclude JavaScript from the Spectare script tag. By default, Spectare loads asynchronously so it should not appear on any defer lists, but if you have aggressive JS optimisation turned on, check that spectare.js is excluded from delay settings.
For Cloudflare users, Spectare is compatible with Cloudflare's Rocket Loader. If you see empty slots after enabling Rocket Loader, add spectare.js to the Rocket Loader exclusion list in your Cloudflare dashboard.
The Spectare slot height reservation (via CSS) works before JavaScript runs, so the layout holds its shape even while the caching plugin serves a fully cached HTML page.
